Yes. Rockstar has confirmed that Grand Theft Auto VI is set in Vice City and the surrounding fictional state of Leonida. That makes it the first mainline Grand Theft Auto to return to Vice City since 2002. How much of the state is playable, and which other towns appear, is not yet announced by Rockstar.

Rockstar's own announcement and trailer material place the game in Vice City, within the state of Leonida. Both names come from Rockstar, not from rumour.
Leonida is a fictional state in the style Rockstar has always used — recognisably inspired by a real place without being it. Vice City sits inside that state rather than standing alone.
What is still unknown about the setting
The playable boundary of Leonida is not announced. Neither is the number of named towns, whether rural areas are fully explorable, or how the world compares in scale to previous games.
